Urbanus Joseph Kioko studied theology at the Kilimambogo Seminar in Kenya (1949-1953) and philosophy at the Kibosho Seminar in Tanzania (1953-1955). He was then a teacher at the Kiserian Seminar in Tanzania and from 1957 to 1960 at the Morogoro Seminar. He was ordained a deacon in 1960, and on January 8, 1961 he received the priestly ordination. He then worked in pastoral care, and in 1966 he changed to Apostolic Vicar after Machakos. In 1967/1968 further studies led him to Italy and England. In 1969/1970 he was a rain of the St. Joseph priestly seminar in Mwingi.
In 1973 he was appointed by Paul VI. the second bishop of the Machakos bishopric. On October 7, 1973, the Bishop gave him the Archbishop of Nairobi Maurice Michael Cardinal Otunga; Co-conspirators were the Archbishop of Kitui William Dunne and the then Bishop of Nakuru and predecessor Raphael S. Ndingi Mwana'a Nzeki.
