AS04

AS04 is an adjuvant used as a pharmaceutical adjuvant in vaccine production. It consists of a complex consisting of an aluminum salt and monophosphoryl lipid A (MPL). It is discussed that the MPL activates the cytotoxic T-cells in AS04 via an activation of the Toll-like receptor TLR4 without activating further inflammatory cells. This results in an increased immune response, which helps to save vaccine antigens. Currently, AS04 (MPL adsorbed to water-containing aluminum hydroxide) is used in the HPV vaccine Cervarix, variant AS04C (MPL adsorbed on aluminum phosphate) in the hepatitis B vaccine Fendrix.
