Duncan Theodore Tsoke studied philosophy at the Priestly Seminary of Hammanskraal in Pretoria and Catholic Theology at the National Seminary of St. John Maria Vianney. On 2 December 1995 he received the sacrament of priestly ordination for the archbishopric of Johannesburg.
He worked as a diocesan youth counselor until 1999 as a diocesan youth counselor. From 1999 to 2000 he was a lecturer for spirituality in the priestly seminary, and subsequently bishop's vicar for evangelism until 2009, and at the same time in the vocational pontoral. Since 2009 he has been Vicar General of the Archdiocese of Johannesburg. After further studies, he earned a degree in ecclesiastical asset management in Nairobi in 2010. On February 6, 2016, Pope Franziskus appointed him as the titular bishop of Horrea Coelia and the auxiliary bishop of Johannesburg. The Archbishop of Johannesburg, Buti Joseph Tlhagale OMI, gave him the bishop's ordination on 30 April of that year.
