Müller-Molinari received her vocal training in Munich with Felicie Hüni-Mihaczek, later with Friedel Becker-Brill in Wuppertal. In 1972 she received her first engagement at the Staatstheater Saarbrücken. Finally, she went to Italy to improve her vocal power with Maria Teresa Pediconi and Giulietta Simionato.

In 1975, Müller-Molinari made his debut in Ravels L'enfant et les sortilèges on La Scala in Milan. In the following years she performed as a guest on numerous stages in Italy as well as at the Opéra National de Lorraine in Nancy and Opera Ireland. At the Salzburger Festspiele she sang several times under the direction of Herbert von Karajan: from 1983 to 1984 the role of the Annina in Richard Strauss' Rosenkavalier, in 1986 the title role in Bizet's Carmen.
